Fluo-3
Fluo-3 absorbs at 506 nm and emits at 526 nm when bound to calcium. It is one of the most widely used fluorescent indicators because it can be optimally excited by the commonlyused 488 argon-ion laser line and its relatively long wavelength emission reduces interference from background fluorescence. Unlike Fura-2 and Indo-1, neither the excitation nor the emission maximum of the dye shifts significantly before and after Ca2+ binding. As a result, the fluorometric ratioing technique is not applicable to Fluo-3. Fluo-3 is essentially nonfluorescent without Ca2+ present, but the fluorescence increases at least 40 times on Ca2+ binding. Because Fluo-3 binds Ca2+ more weakly (higher Kd) than Fura-2 and Indo-1, it is more useful for measuring high transient Ca2+ concentration during Ca2+ spikes.
Like other AM esters, Fluo-3 AM ester is relatively unstable when frequently exposed to moisture.
