Indo-1
Indo-1 is a UV-excitable fluorescent Ca2+ indicator that can be optimally excited by the 351-356 nm spectral lines of the argon-ion laser. The emission maximum of the indicator shifts from ~475 nm in Ca2+-free medium to ~400 nm in Ca2+-saturated medium (Figure 1.9). Ca2+ concentration can thus be determined by ratioing the fluorescence intensities at the two wavelengths. As with Fura-2, this ratioing technique avoids problems associated with uneven dye distribution, cell or tissue thickness and photobleaching. Indo-1 has been widely used in flow cytometry studies.
