Amine-reactive Dyes and Related Reagents
The most common reagents for labeling amines are succinimidyl esters (SE). SE reagents can readily react with primary and secondary aliphatic amines to form a highly stable amide bond. Labeling of water-soluble amine-containing compounds such as peptides, proteins and oligonucleotides are typically carried out in an aqueous buffer with a pH of 8.0 to 9.0 Amine labeling can also be carried out in an organic solvent for compounds that have low water solubility. Biotium offers a a large number of dyes and haptens (biotin) with a SE group. Our high quality (>98% purity) single isomer rhodamine and fluorescein SE products are excellent for nucleic acid sequencing and other applications where a single species product is desired. In addition, we have several products that have a long PEG spacer arm between the SE group and the dye. These products offer increased water-solubility and on the other hand reduced steric effect by the dye on the molecule to be labeled.
Finally, other amine-reactive dyes such as fluorescent dyes with sulfonyl chloride, isothiocyanate and dichlorotriazine are also available from Biotium. These reactive dyes, however, are usually not as widely used as the SE products, either because they are not as convenient to use or because the linkage of the labeled products are not as stable.
