RedDot^TM2 is a far-red cell membrane-impermeable nuclear dye similar to Draq7^TM. The dye is ideal for specifically staining the nuclei of dead cells with minimal cytoplasmic RNA staining. RedDot^TM2 provides excellent nuclear counterstaining in fixed and permeabilized cells and tissue sections without the need for RNase treatment to remove cytoplasmic RNA. The dye is highly thermostable and photostable, providing convenient handling and ideal for demanding applications such as confocal microscopy.

HeLa cells stained with mouse anti-Golgin 97 (Golgi marker) detected with CF555 goat anti-mouse IgG (cyan) and rabbit anti-COXIV (mitochondrial marker) detected with CF488A goat anti-rabbit IgG (green). Actin filaments are stained with CF405 phalloidin (blue), and nuclei are counterstained with RedDot2 (red).

Formaldehyde fixed and detergent permeabilized HeLa cells stained with 1X RedDot2 (top) or 3 uM Draq7 (bottom) in PBS for 10 minutes. RedDot2 staining is highly selective for the nucleus. In contrast, Draq7 stains the nucleus and cytoplasm unless a separate blocking step is performed. Actin filaments are stained with CF488A phalloidin (green).
