The new universal antigen retrieval buffer allows researchers to recover immunostaining targets made unavailable by tissue preservation.
Fremont, CA, May 21, 2020: Biotium will release a new product, AntiFix™ Universal Antigen Retrieval Buffer, to benefit immunohistochemistry researchers working on tissue samples. AntiFix™ is designed for heat-induced epitope retrieval (HIER) of formalin-fixed, paraffin-embedded (FFPE) tissues. The buffer uses a proprietary reagent that catalyzes the reversal of formaldehyde cross-links in FFPE tissue to restore antigen reactivity. AntiFix™ Universal Antigen Retrieval Buffer can be used at neutral pH and offers a new way for researchers to simplify and optimize HIER providing excellent reproducibility and efficiency.
Fixation of tissue with formalin (formaldehyde) preserves tissue morphology. However, the cross-linking of proteins by formaldehyde can modify antibody binding sites or epitopes, so they are no longer detectable by immunohistochemistry. Traditionally, epitope reactivity is recovered using protease treatment or by heating slides in either low pH citrate buffer or high pH Tris buffer. The ideal conditions for unmasking varies for different epitopes, requiring a tedious troubleshooting process involving multiple buffers to be used for different targets. AntiFix™ Universal Antigen Retrieval Buffer streamlines this process with a formulation that is optimized to work with a wide variety of immunohistochemistry targets, reducing the need for buffer optimization during HIER protocols. AntiFix™ Universal Antigen Retrieval Buffer has been validated in immunofluorescence staining of FFPE tissue with Biotium’s next-generation fluorescent CF® dyes using HRP-mediated tyramide signal amplification.
Biotium is an industry leader in providing fluorescently labeled antibodies and cell stains for fluorescence microscopy. By including accessories such as AntiFix™ Universal Antigen Retrieval Buffer, mounting media, and autofluorescence quenchers in their product line, Biotium continues to support researchers in all aspects of their workflow.
