Unique hardset antifade that quenches lipofuscin autofluorescence. Choose with mounting medium with DAPI, far-red NucSpot® 640, or without nuclear stain.
This unique hardset antifade mounting medium quenches lipofuscin autofluorescence, for one-step mounting, quenching, and optional nuclear counterstaining of fluorescently-stained tissue sections.
TrueBlack® Lipofuscin Quenchers are designed to quench lipofuscin autofluorescence, a common source of background in human and aged animal tissues such as brain and retina. They also may reduce background from other sources like red blood cells or extracellular matrix. EverBrite TrueBlack® Hardset Mounting Medium performs lipofuscin quenching and mounting in a single step, for greater convenience. The mounting medium substantially reduces lipofuscin fluorescence after mounting, and quenching continues to improve as the medium cures and hardens overnight. Unlike VECTASHIELD® Mounting Medium, EverBrite TrueBlack® is compatible with cyanine-based fluorophores and is optimally formulated for use with Biotium’s CF® Dyes.
Note: EverBrite TrueBlack® Hardset may reduce specific fluorescence; antibody or probe concentration may require optimization for use with the mounting medium.
EverBrite TrueBlack® is available with or without DAPI, or with Biotium’s novel NucSpot® nuclear-specific counterstains. NucSpot® 640 is a far-red dye for the Cy®5 channel. Using a far-red nuclear stain can avoid problems of fluorescence cross-talk and UV photoconversion with traditional dyes like DAPI. Note: NucSpot® dyes also show dim blue fluorescence with the DAPI filter set, and should be tested for suitability before using with blue probes, especially for epifluorescence microscopy.

TrueBlack® Lipofuscin Autofluorescence Quencher, 20X in DMF (Cat. No. 23007) is the original formulation. TrueBlack® Lipofuscin Autofluorescence Quencher, 30X in DMSO (Cat. No. 23011) is a newer, more concentrated formulation in DMSO, which is a less toxic solvent than DMF. The quencher stock solution formulation does not affect the performance in staining.
TrueBlack® Lipofuscin Autofluorescence Quencher and TrueBlack® Plus Lipofuscin Autofluorescence Quencher were not designed for colorimetric detection, but they will stain lipofuscin black and be visible in light microscopy.
TrueBlack® Lipofuscin Autofluorescence Quencher (Cat. No. 23007, 23011) and TrueBlack® Plus Lipofuscin Autofluorescence Quencher (Cat. No. 23014) are designed to reduce autofluorescence from lipofuscin in tissue samples such as mouse and human brains and retina. While TrueBlack® Lipofuscin Quenchers have been reported to reduce autofluorescence from other sources, such as collagen, elastin, red blood cells, and general background fluorescence, they are not as effective at quenching these types of autofluorescence as for lipofuscin autofluorescence. However, they may improve background from a variety of sources in different experimental systems.

EverBrite™ mounting medium does show high background fluorescence with violet excitation. The background is more prominent when imaged using epifluorescence using UV excitation and a DAPI filter cube, but is generally quite low in confocal imaging using 405 nm laser.
CoverGrip™ is designed to replace nail polish for permanent sealing of wet-mounted coverslips. It can be removed by soaking slides in buffer (see the product information sheet for details). CoverGrip™ is not designed to create a removable temporary seal for hybridization.
Yes, EverBrite™ mounting media are compatible with a wide range of dyes. Unlike VectaShield®, EverBrite™ won’t interfere with cyanine-based fluorophores like Alexa Fluor® 647, Cy®3, or Cy®5.
