What is the refractive index of EverBrite™ Mounting Medium?

EverBrite™ Mounting Medium (23001) and EverBrite™ Mounting Medium with DAPI (23002) both have a refractive index of 1.46.  EverBrite™ Hardset Mounting Medium (23003) and EverBrite™ Hardset Mounting Medium with DAPI (23004) both have an initial refractive index of 1.38, which increases to 1.42 after 24 hours of curing, and 1.46 after 4 days, after which it remains constant

Are the EverBrite™ Mounting Media, wetset and hardset, compatible with Alexa Fluor® dyes or Cy® dyes?

Yes, EverBrite™ mounting media are compatible with a wide range of dyes. Unlike VectaShield®, EverBrite™ won’t interfere with cyanine-based fluorophores like Alexa Fluor® 647, Cy®3, or Cy®5.

Can I remove EverBrite™ Mounting Medium from my samples after mounting?

EverBrite™ wet-set mounting medium is an aqueous, glycerol-containing medium. It can be removed from samples by washing several times with PBS or other buffer. However, washing with PBS will not remove DAPI staining.

For coverslips mounted on slides with EverBrite™ Hardset or EverBrite™ wet-set medium, the slides can be soaked in PBS in a slide jar for several hours with gentle agitation until the coverslip slides off (avoid prying or pulling the coverslip, which could damage the sample). Then wash the slide several times to remove any remaining mounting medium. If the coverslips have been sealed with CoverGrip™ Coverslip Sealant, soaking in PBS will remove the sealant as well. Other methods may be required to remove other types of coverslip sealants.

Biotium’s EverBrite™ mounting medium is a revolutionary antifade mounting medium optimally formulated for preserving fluorescence of our CF® dyes and other fluorochromes for your immunofluorescence (IF) imaging applications.

  • Antifade mounting medium optimally formulated to prevent photobleaching of fluorophores
  • Refractive index is well-matched to that of coverslip glass and immersion oil
  • Available with or without DAPI

EverBrite™ provides a high level of protection from photobleaching to prolong fluorescence over a wide range of wavelengths. EverBrite™ Mounting Medium has a refractive index of 1.46, which is well-matched to the refractive indices of coverslip glass and immersion oil (1.5). The mounting medium remains liquid and permits immediate imaging of slides and tissues after sealing the cover slip edges with nail polish or Biotium’s CoverGrip™ Coverslip Sealant. After sealing, the slides can be stored at 4°C for a year or longer. EverBrite is available with or without DAPI for nuclear counterstaining. Unlike VectaShield®, EverBrite™ is compatible with cyanine-based fluorophores like Cy® dyes.

Photostability of Cy®5 staining in HeLa cells with various mounting media. A major advantage of EverBrite™ mounting medium is that, unlike Vectashield®, it does not decrease the fluorescence of cyanine-based fluorophores. Fluorescence values for Cy®5 in Vectashield® media are normalized to PBS time t0 to illustrate the drop in fluorescence of cyanine dyes caused by Vectashield®.
