A magnetic bead-based kit for superior RNA extraction from FFPE samples. Features proprietary technology for enhanced removal of formaldehyde-induced damage, providing RNA with higher yield and greater amplifiability.
Formalin-fixed tissue samples are a challenge for RNA extraction, often resulting in low yields and poor performance in subsequent steps. Most existing methods rely on heat to remove crosslinks and adducts, which is only partially effective and leads to additional fragmentation of labile nucleic acids. Powered by proprietary CAT5™ catalytic technology, the RNAstorm™ FFPE RNA Extraction Kits enhance the removal of formaldehyde-induced damage. The extracted RNA has higher yield, quality, and greater amplifiability that is more optimal for next-generation sequencing, qPCR, microarray, or other gene expression analysis.

Comparison of RNA recovered from human kidney FFPE sections using the CELLDATA RNAstorm™ 2.0 FFPE RNA Extraction Kit using MagBeads or spin columns. RNA was isolated from four 10 um FFPE sections per sample using duplicate samples for each kit. Eluted RNA (15 uL per lane) was analyzed on a 1.5% EMBER™ Ultra RNA agarose gel and imaged on Biotium’s Gel-Bright® Laser Diode Gel Illuminator. The RNAstorm™ 2.0 FFPE MagBead FFPE RNA Kit yielded RNA with comparable recovery and RNA size profiles as the RNAstorm™ 2.0 kit with spin columns. Ladder: NEB Low Range RNA Ladder. Significant improvements in RNA yield and quality (as measured by amount of amplifiable RNA) are seen using a prototype of the RNAstorm™ kit featuring CAT5™ technology on FFPE samples from various tissues and with collection dates ranging from 1976 to 2015 compared to a competitor kit.

Increased DV200 values are observed for RNA extracted using the RNAstorm™ kit relative to a popular commercial kit. The DV200 represents percentage of RNA with length greater than 200 nt, as measured using an Agilent Bioanalyzer RNA 6000 nano kit.

The CELLDATA RNAstorm™ 2.0 FFPE RNA Extraction Kit (Cat. No. CD506) and CELLDATA DNAstorm™ 2.0 FFPE DNA Extraction Kit (Cat. No. CD507) contain a Dewaxing Solution that was reformulated for IP considerations. These kits are direct replacements for the discontinued CELLDATA RNAstorm™ or DNAstorm™ FFPE Extraction Kits (CD501, CD502). The kits continue to utilize Biotium’s proprietary CAT5™ catalytic technology for efficient reversal of formaldehyde crosslinks.
The maximum capacity of the spin columns in the kit is similar to a standard miniprep column, about 20 ug of DNA. However, the expected yield from FFPE extraction is much lower, it is very rare to get more than 1-2 ug of DNA per prep.
Yes, the RNAStorm™ and DNAStorm™ FFPE kits may be used sequentially. The steps below will allow the protocol to be adapted to extract both RNA and DNA from one sample. You can also download this information in an app note.
Begin by extracting the sample according to the RNAstorm™ kit protocol with the following modifications:
Note: the initial incubation period can be adjusted depending on relative DNA and RNA yields. If the RNA yield is high but the DNA yield is low, reduce the incubation time in step 3 (no less than 15 mins). If the DNA yield is good but the RNA yield is low, increase the incubation time in step 3 (no more than 2 hours).
