What are Single-Domain Antibodies?

Single-domain antibodies (SdAbs), also known as VHH domains, are small antigen-binding fragments derived from the variable domain of heavy chain antibodies (HCAbs) that are found in camelids such as alpacas, llamas, and camels.

Conventional antibodies, such as mouse, rabbit, or rat IgGs, are heterodimers composed of two heavy and two light chains. Their antigen-binding site forms through the interaction of variable domains from both chains. In contrast, camelid HCAbs (IgG2 and IgG3 subclasses) lack light chains and the CH1 domain, relying on a single variable domain for antigen recognition.
